Der Film behandelt das Thema der Bestimmung von Lebewesen anhand des dichotomischen Bestimmungsschlüssels. Dieser fußt auf der taxonomischen Einordnung, bildet eine Systematik und bezieht sich auf Entscheidungen, denen jeweils ein zweigliedriger Entscheidungsbaum zugrunde liegt.
Aufgebaut ist der Film in die Abschnitte der Entwicklung der Systematik und des Systems des dichotomen Bestimmungsschlüssels, der Beschreibung von Irrtümern und Fehlern, die bei einer Bestimmung auftreten können und einem praktischen Beispiel der Bestimmung einer Pflanze anhand von Merkmalen.
